Participate in a project that aims to decrease colorectal cancer death (CTMM DeCoDe).
The post-doc will be responsible for the chemistry to develop molecular imaging reagents for two different imaging modalities, MRI and PET.
At the life science facilities of Philips Research in Eindhoven the 19F base MRI contrast agents will be developed. The 19F probes are based on micro-capsules containing a perfluoro-compound. The micro-capsules will be targeted against several bio-markers and evaluated in in vivo models.
The efficacy of these probes will be assessed by multimodality imaging, e.g. additional SPECT/CT as well as optical (fluorescent) imaging. For this purpose perfluoro emulsions stabilized with amphiphilic compounds will be synthesized, and ligands to target disease related molecular markers will be conjugated for molecular imaging applications. Experimental work on molecular imaging of colon cancer cells in vitro (cell lines), ex vivo (resection material), and in vivo (mouse models) will be carried out mainly at the VUmc (Amsterdam) and partly at the Life Science Facility of Philips Research.
At the department of Nuclear Medicine & PET Research new selective radiopharmaceuticals for PET imaging of AURKA will be developed. New radiopharmaceuticals will be radiolabeled with Carbon-11 based on known selective AURKA inhibitors and evaluated in vitro and in vivo models of disease. After being successful in these in vivo models, GMP compliant synthesis and validation in humans is foreseen.
The candidate will be appointed at the VUmc. The work will be carried out in close collaboration with all partners, in Amsterdam and in Eindhoven.

Gestelde eisen
The candidate should have a PhD in radiopharmaceutical sciences, medicinal chemistry, organic chemistry or pharmaceutical sciences with focus on organic chemistry, preferably with expertise in colloid chemistry. Prior experience with radiochemistry is a plus. The project requires experimental work and broad interests in adjacent areas, such as imaging technologies, cell cultures, and preclinical studies.

Afdeling nuclear medicine and PET research in collaboration with phillips research eindhoven Colorectal cancer (CRC) is the third highest cause of cancer-related mortality in the western world. Globally, the annual economic burden for CRC is estimated to be in the range of $US 14 - 22 billion. Early detection and staging strategies, together with personalized therapies based upon individual tumor characteristics, are generally considered as the best way to improve patient outcomes and reduce the economic burden of CRC. The CTMM-funded DeCoDe project aims to decrease colorectal cancer death (CTMM DeCoDe). It consists of a consortium of five university medical centres and eight industrial partners in The Netherlands, that jointly work in the fields of early molecular detection, personalized therapies and molecular imaging of colorectal cancer. Within the collaborative molecular imaging efforts between VU University Medical Center Amsterdam and Philips Research Eindhoven there is a vacancy for a post-doc for a period of 2 years for generation of MRI contrast reagents and PET ligands. MRI based Molecular Imaging methods are being developed for CRC screening purposes, based on fluorine contrast agents. Selective radiopharmaceuticals for Aurora Kinase A (AURKA) need to be developed as a tool to study this enzyme by Positron Emission Tomography CRC metastasis. The radiopharmaceutical chemistry section (department of Nuclear Medicine & PET Research) consists of 5 staff chemist, 7 PhD's, 5 post-doc's and 15 research technicians. It has an extensive experience in the development of radiopharmaceuticals. They work closely together with physicists, biologists and physicians in this research. This typical multidisciplinary environment guarantees a broad area of research for a Post-doc, where ideas and challenges are met at the interface of medicine, chemistry and biology. At Philips Research the candidate will be a member of bio-medical engineering group and will have access to the chemical and radio-chemical facilities.
